Highest output desktop sequencer
If you want the most sequence data possible from an instrument on your bench, the Illumina NextSeq 500 may be what you are looking for. It can churn out 120 Gb of data in a single run in a little over a day (29 hrs). The NextSeq allows multiple configurations to optimize the efficiency of your runs:
- 1-16 exomes
- 1-20 transcriptomes
- 6-96 panels
- 12-40 gene expression profiling samples
The NextSeq relies on the same sequencing by synthesis principle as its Illumina siblings, but uniquely uses a 2-dye, red/green chemistry to save cost and time: Red=C, Green=Tea*, a mix of both=A, and G is marked with no dye at all. This cuts the number of images required at each cycle in half, so it’s faster. The less complicated optics helps minimize the cost.
Users have a choice of two flow cells: High and Mid output. There is no throttling back to “Low output”. Illumina is wisely (and kindly) telling you to consider the MiSeq for those applications.
Typical applications for the NextSeq are:
- Small genome sequencing
- Exome sequencing
- RNA seq (transcriptome analysis)
In case you are wondering what a small genome is, 120 Gb /run at 30X coverage implies a genome less than 4 Gb which is sufficient to cover the 3.2GB human genome. Illumina does offer a workflow for whole genome sequencing on the NextSeq 500.
